抗戰期間，日軍占領東北三省，少女翠英以賣唱維生，與愛國青年周凌雲相戀，卻被迫忍辱嫁給垂涎她的漢奸陳兆群。數年後，陳兆群任偽政府特務機關首腦，為全力追捕國民政府特務「天字第一號」而焦頭爛額。周凌雲作為其外甥，被國民政府吸收後潛入陳家任偽職，卻驚覺當年戀人已成為姑丈之續絃。情感受創之餘，他另起情緣，投身間諜行動，誓圖抗日復國。殊不知，原來叱咤風雲的抗日頭號間諜「天字第一號」竟是⋯⋯。

本片由金馬影帝柯俊雄與「千面女郎」白虹聯合主演，翻拍自1946年轟動上海、屠光啟執導的同名作品，為本土首部自製台語諜報片，不僅創下票房紀錄，更開啟台語間諜片風潮，前後衍生五集系列作品，盛況轟動一時，成為一代觀眾的共同記憶。

During the Sino-Japanese War, songstress Tshui-ing falls in love with young patriot Ling-hun, but is forced to marry an older collaborator serving the Japanese occupation. Years later, Ling-hun is recruited by the Nationalists and infiltrates his uncle's household, only to discover his former lover is now his uncle's wife. As the puppet regime intensifies its hunt for the elusive resistance agent "Special Agent No. 1," Ling-hun devotes himself to the anti-Japanese cause, while the legendary spy's true identity slowly comes to light.
