抗戰期間，日軍占領東北三省，以賣唱為生的少女翠英與青年周凌雲相戀，卻忍辱嫁給垂涎於她的漢奸陳兆群。數年後，陳兆群為偽政府主持特務機關，為了拘捕國民政府特務「天字第一號」焦頭爛額。周凌雲身為陳兆群的外甥，被國民政府吸收後在陳家擔任偽職，才發現當年戀人已成為姑丈的續絃。神傷之餘，他展開新戀情、全心投入間諜任務，盼望抗日復國。萬萬沒想到，原來叱咤風雲的抗日頭號間諜「天字第一號」竟是⋯⋯

本片由金馬影帝柯俊雄、六○年代「千面女郎」巨星白虹合作主演，翻拍自1945年轟動上海的同名電影，是首部本土自製的台語諜報片。爾虞我詐的劇情、面貌多變的「女間諜」角色，讓《天字第一號》彷如臺灣版007系列電影，創下臺灣電影票房紀錄，也開啟台語間諜類型片數年風潮，連續拍了五集，轟動一時，成為世代記憶。

During the Sino-Japanese War, Cuiying flees with her father from the Japanese occupation. She meets a young man named Lingyun on the way, but soon bids farewell to him. After years of exile, they reunite in Shanghai and fall in love. Yet Cuiying chooses to leave Lingyun and marries his uncle Zhaoqun, a pro-Japanese collaborator. Shocked and heartbroken, Lingyun leaves China to study abroad. Meanwhile, “Special Agent 001” leads the sabotage against the Japanese, bringing tremendous pressure to Zhaoqun. After Lingyun returns to Shanghai, Zhaoqun urges him to marry his daughter Aili, who disagrees with her father’s rapport with the Japanese and secretly aids the resistance movement. While a struggle about love and patriotism ensues in this family, Special Agent 001 continues the sabotage and the identity of the agent finally reveals…
